The dawn of
sequencing:
Protein
sequencing-
The earliest techniques were based
on methods for separation of proteins and
peptides, coupled with methods for identification and quantification of amino acids. Initially,
the majority of protein sequences were
obtained by the manual process of sequential Edman degradation - dansylation. Advanced in mass
spectrometry allowed significant
progress.
Nucleic acid
sequencing-
The first technique were only
applicable to RNA. Fast, and efficient DNA
sequencing had emerged using chain termination (Sanger) and chemical degradation (Maxam and
Gilbert) approaches.
